#1 - Hepatic encephalopathy secondary to poisoning by Tephrosia cinerea in sheep

Abstract in English:

In the semiarid region of northeastern Brazil, Tephrosia cinerea causes periacinar hepatic fibrosis in sheep with severe ascites and, occasionally, nervous signs. Sixteen sheep from six outbreaks of T. cinerea poisoning were studied. All sheep had histologic lesion of periacinar fibrosis and six showed, in the brain, vacuolization (spongy degeneration) of the white matter and junction between grey and white matter and presence of Alzheimer type II astrocytes in the grey matter. The disease was produced experimentally in two sheep, that presented porto-sistemic shunts and similar histologic lesions as those observed in the spontaneous cases. Immunohistochemistry revealed weak labelling with anti-GFAP antibodies suggesting a degenerative alteration of astrocytes with accumulation of dense bodies and reduction of the GFAP. There was strong labelling with anti-S100 antibodies suggesting cellular reactivity with proliferation of mitochondria and endoplasmatic reticulum. Such alterations are characteristic of the effects caused by ammonia on the astrocytes. It is concluded that in poisoning by T. cinerea nervous signs due to hepatic encephalopathy occur in some sheep.

A intoxicação por Tephrosia cinerea causa fibrose hepática periacinar em ovinos na região semiárida do Nordeste, com quadro clínico de ascite acentuada, e, ocasionalmente, com sinais neurológicos. Neste trabalho foram estudadas 16 ovinos em 6 surtos de intoxicação por T. cinerea. Todos os ovinos apresentaram lesões histológicas de fibrose periacinar e seis apresentaram, no encéfalo, vacuolização da substância branca e da junção entre a substância branca e a cinzenta com presença de astrócitos de Alzheimer tipo II na substância cinzenta. A doença foi reproduzida experimentalmente em dois ovinos que apresentaram ascite, desvios vasculares (shunts) porto-sistêmicos e sinais nervosos com lesões histológicas semelhantes a dos casos espontâneos. Na técnica de imuno-histoquímica houve marcação fraca ou ausente do citoplasma astrocitário para o anticorpo anti-GFAP em seis ovinos evidenciando uma alteração degenerativa, em que os astrócitos acumulam corpos densos e reduzem o volume de GFAP. Houve marcação positiva para o anticorpo anti-S100 em oito ovinos, incluindo os dois ovinos experimentais o que sugere reatividade celular, com proliferação mitocondrial e de retículo endoplasmático liso. Estas alterações são caraterísticas dos efeitos da amônia nos astrócitos. Conclui-se que na intoxicação por T. cinerea em alguns ovinos ocorrem sinais nervosos em consequência da encefalopatia hepática.

E-mail: angelicawouters@yahoo.com.br Senecio spp. poisoning is an important cause of illness and death of cattle in Rio Grande do Sul, Brazil, and is often manifested by neurologic clinical signs and histological brain changes. Histological evaluation was performed on liver and brain samples of ten cattle naturally poisoned by Senecio sp. Samples of cerebrum, brainstem and cerebellum were stained with periodic acid-Schiff (PAS), and immunohistochemistry was carried out, employing anti-fibrillary acidic protein (GFAP), anti-S100 protein and anti-vimentin antibodies. The histological finding in the brain included mild to severe vacuolation in the white matter and the junction of gray and white matter, characterized as spongy degeneration. Histochemical and immunohistochemical staining revealed no significant findings compared with the brains of eleven adult cattle without liver and/or brain changes used as controls.

Foi efetuada avaliação histológica em fígado e encéfalo de dez bovinos naturalmente intoxicados por Senecio sp. e foram realizadas as técnicas de histoquímica de ácido periódico de Schiff e de imuno-histoquímica, com emprego dos anticorpos anti-proteína fibrilar ácida (GFAP), anti-proteína S100 e anti-vimentina em amostras de telencéfalo, tronco encefálico e cerebelo. Na histologia do SNC observou-se vacuolização discreta a acentuada em substância branca e/ou junção entre substância branca e cinzenta, caracterizada como degeneração esponjosa. Na avaliação histoquímica e imuno-histoquímica não houve diferenças significativas em relação aos achados em encéfalo de onze bovinos controle, adultos, sem alterações hepáticas e/ou encefálicas, avaliados pelas mesmas técnicas.


#3 - Degeneração esponjosa no sistema nervoso central de bezerros da raça Sindhi

E-mail franklin.riet@pesquisador.cnpq.br Spongy degeneration (status spongiosus) includes many histological alterations characterized by vacuoles in the neuropile in different regions of the central nervous system (CNS). The vacuolization can be caused by intramyelinic edema, as in maple syrup urine disease and some toxic diseases, or by astrocytic edema, as in citrullinemia. This paper reports a spongy degeneration in two Sindhi calves, a female and a male. Both calves were offspring from the same bull, and the herd had high degree of inbreeding. The female calf was born normal, but progressive nervous signs were observed since 2 months old. The male calf had progressive nervous signs since it was born. Both calves were euthanized with severe nervous signs when 4 months old. At necropsy the liver was pale in the male calf. Histological alterations were mild to accentuated vacuolization, mainly in the deep cerebral cortex, cerebellar white matter, grey matter of the spinal cord, internal capsule, and brain stem. Fatty degeneration was observed in the liver of both animals. On electron microscopy it was found that the status spongiosus was caused by astrocytic edema. It is suggested that the disease is caused by a metabolic hereditary error.

#4 - Occurrence of outbreaks of Senecio spp. (Compositae) poisoning in cattle in Rio Grande do Sul, Brazil

Abstract in English:

Three outbreaks of Senecio spp. poisoning in cattle in Rio Grande do Sul, Brazil, are described. At least 13 bovines died due to the intoxication in the counties of Santo Augusto, São Sepé and Júlio de Castilhos. Clinical signs included depression, abdominal straining, abdominal pain, loss of appetite and nervous signs such as incoordination of gait, circling and agressivity. At necropsy the most consistent lesions were edema of the body cavities, mesentery, abomasal folds, gall bladder wall and subcutaneous tissue of the ventral parts of the body, fibrous and hard livers and firm and white spotted kidneys. Microscopic lesions included different degrees of fibrous proliferation and megalocytosis in the tiver and kidney with ductal hyperplasia in the former a protein loosing glomerulopathy in the latter. Vacuolar and hyalin degeneration were seen in the myocardial fibers and spongy degeneration in the white matter of the brain. Associated with the pastures where the affected animals had been grazing, abundant quantities of two species of Senecio were found, namely S. brasiliensis and S. selloi. No sheep were grazing at the sarne pastures where cattle tosses occurred.

São descritos três surtos de intoxicação por Senecio spp. em bovinos no Rio Grande do Sul. Os surtos ocorreram nos municípios de Santo Augusto, São Sepé e Júlio de Castilhos, causando pelo menos 13 mortes. Sinais clínicos dos animais afetados incluíam depressão, tenesmo, dor abdominal, anorexia e sinais nervosos como incoordenação motora, andar· em círculos e agressividade. As lesões mais consistentemente vistas na necropsia foram edemas cavitários, do mesentério, das pregas do abomaso, da parede da vesícula biliar, do tecido subcutâneo das partes ventrais do organismo e endurecimento do fígado e rim por proliferação fibrosa. O rim mostrava ainda manchas brancas no córtex. As lesões microscópicas consistiam de diferentes graus de proliferação fibrosa e megalocitose no fígado e rim, com hiperplasia ductal no primeiro e glomerulopatia com perda de proteína neste último. Degeneração vacuolar e hialina de fibras cardíacas e degeneração esponjosa da substância branca do encéfalo foram também observadas. As pastagens, onde os animais afetados tinham estado, encontravam-se intensamente infestadas por duas espécies de Senecio, S. brasiliensis e S. selloi. Nessas pastagens não havia ovinos.
